KOSTAS VERDELIS: Study Of Mineral And Matrix Maturation In Dentin (Under the direction of Adele L. Boskey and J. Timothy Wright) Spectroscopic analysis was used to study the patterns of changes in the mineral and matrix properties of dentin during maturation of the tissue. Fourier Transform Infrared Imaging (FTIRI) analyses on undecalcified semi-thin sections from fetal bovine incisors and developing mouse molars were performed. In addition, fetal bovine microdissected mantle and circumpulpal dentin specimens of successive tissue age were analyzed by Fourier Transform Infrared (FTIR) analysis and by amino acid and matrix phosphate assays. In the initial studies, the formation of mantle and circumpulpal dentin as two distinct dentin compartments in the developing fetal incisors was established through analysis of distribution of mineral:matrix and mineral crystallinity values. Changes in the mineral:matrix, mineral crystallinity, acidic phosphate substitution and carbonate substitution in the mineral of mantle and circumpulpal dentin during maturation were subsequently quantitatively analyzed from FTIRI results. In this study, separate patterns of changes in mineral properties were found for mantle and circumpulpal dentin, in terms of initial and final levels and rates of increase or decrease of mineral properties values. Osteocalcin Mediates Biomineralization during Osteogenic Maturation in Human Mesenchymal Stromal Cells

There is a growing interest in cell therapies using mesenchymal stromal cells (MSCs) for repairing bone defects. MSCs have the ability to differentiate into osteoprogenitors and osteoblasts as well as to form calcified bone matrix. However, the molecular mechanisms governing mineralization during osteogenic differentiation remain unclear. Non-collagenous proteins in the extracellular matrix are...

Relationships between protein and mineral during enamel development in normal and genetically altered mice.

The purpose of this study was to quantify and compare the amounts of volatiles (mostly protein) and mineral present in developing incisor enamel in normal mice and in those genetically engineered for absence of intact enamelin, ameloblastin, matrix metalloproteinase 20 (MMP20) or kallikrein-related peptidase 4 (KLK4). Data indicated that all mice showed peaks in the gross weight of volatiles an...

Effect of kallikrein 4 loss on enamel mineralization: comparison with mice lacking matrix metalloproteinase 20.

Enamel formation depends on a triad of tissue-specific matrix proteins (amelogenin, ameloblastin, and enamelin) to help initiate and stabilize progressively elongating, thin mineral ribbons of hydroxyapatite formed during an appositional growth phase. Subsequently, these proteins are eradicated to facilitate lateral expansion of the hydroxyapatite crystallites. The purpose of this study was to ...

Decreased mineral content in MMP-20 null mouse enamel is prominent during the maturation stage.

During enamel development, matrix metalloproteinase-20 (MMP-20, enamelysin) is expressed early during the secretory stage as the enamel thickens, and kallikrein-4 (KLK-4, EMSP1) is expressed later during the maturation stage as the enamel hardens. Thus, we investigated whether the physical properties of the secretory-/maturation-stage MMP-20 null enamel were significantly different from those o...

P-110: Effect of Increasing Amount of Oocyte Secreted Factors on Cumulus Expansion of Bovine Cumulus-Oocyte Complexes

Background: In vitro maturation is a good method to decrease cancer risk of superovulation by gonadotropin hormones. A paracrine effect of oocyte secretions on oocyte developmental competence is under investigation. Apart from oocyte maturation, ovulation in vivo requires a precise control of extracellular matrix modification. Cumulus cells secrete hyaluronan to form a muco-elastic extracellula...
